چکیده

Plasma fibronectin is a component of the fibrin clot, a provisional matrix that arrests bleeding but that can also obstruct flow in blood vessels. The goal of this study was to examine the influence of fibronectin on the formation, structural characteristics and composition of reconstituted fibrin clots. Fibrin clots were formed by adding thrombin to 1 mg/ml, 2 mg/ml or 4 mg/ml fibrinogen supplemented with 0-0.4 mg/ml fibronectin. The formation of fibrin clots with varying amounts of fibronectin was monitored by measuring light absorbance during polymerization. Confocal microscopy of fluorescein conjugated fibrinogen was used to determine the structural characteristics of fibrin-fibronectin clots. The composition of the fibrin-fibronectin clots was determined through electrophoresis and immunoblotting of solublized clots. Fibronectin concentration positively correlated with the initial rate of polymerization and with the steady state light absorbance values in fibrin clots. The effect of fibronectin on fibrin clots was saturable; there was no significant difference in the steady state light absorbance value of fibrin-fibronectin clots formed from 2 mg/ml and 4 mg/ml fibrinogen. At fibrinogen concentration of 2 mg/ml and above, an increase in fibronectin concentration resulted in thinner and denser fibers in the fibrin-fibronectin clots. Electrophoresis and immunoblotting showed that fibronectin bound to fibrin clots and formed covalently bonded high molecular weight multimers. These findings are novel because they correlate results from light absorbance studies to microcopy images and demonstrate an influence of fibronectin on fibrin clot structural characteristics. This data is important in developing therapies that destabilize fibrin clots.

Fibronectin and fibrin gel structure.

Plasma fibronectin is covalently incorporated into alpha-chains of fibrin gels in the presence of Factor XIII activated by thrombin (FXIIIaT) but not by Factor XIII activated by the snake venom enzyme batroxobin (FXIIIaB). FXIIIaB catalyzes introduction of gamma-gamma cross-links in fibrin but cross-linked alpha-chains are not formed. In the presence of FXIIIaT, fibrin gels formed by batroxobin...

Fibronectin provides a conduit for fibroblast transmigration from collagenous stroma into fibrin clot provisional matrix.

After injury, the wound space is filled with a fibrin/fibronectin clot containing growth factors released by platelets and monocytes. In response to these factors, fibroblasts migrate into the fibrin clot and contribute to the formation of granulation tissue. The functional mechanisms allowing fibroblasts to leave the collagenous matrix of normal connective tissue and invade the provisional mat...

Rat hepatocytes in serum-free primary culture elaborate an extensive extracellular matrix containing fibrin and fibronectin

Adult rat hepatocytes cultured on type IV collagen, fibronectin, or laminin and maintained in serum-free medium were examined by indirect immunofluorescence using polyclonal antibodies against extracellular matrix proteins. An extensive fibrillar matrix containing fibronectin and fibrin was detected in all hepatocyte cultures irrespective of the exogenous matrix substratum used to support cell ...

Fibrin formation: effect of calcium ions.

Using laser fluctuation spectroscopy, a technique that measures particle size change in solution, the kinetics of fibrin clot formation from fibrinogen can be studied. With this technique the effect of calcium on the three distinguishable phases of clot formation, (1) proteolysis of fibrinogen, (2) fibrinogen-fibrin monomer complex formation, and (3) fibrin monomer polymerization, were investig...
